Do you have a new iTunes or something…
It could be that the iTunes is not compatiable with the new version of iTunes..??
And has it ever worked before, or is this the first time you’ve tried it??

Try borrowing a friends cable to see if its a faulty connection in yours.
Also look in itunes media settings, you could of accidently turned off the sync, by clicking “I will manage my own music”

no, it would be impossible to be compatibility problem, even if you have the latest version of iTunes with the first generation iPod and even if your iTunes did set to manual, it should’ve start right after you plug your iPod to your computer.
the problem is i think somehow automatic start-up service has been disabled, see on task managers for ipodservices.exe, if it’s not there, then something on your computer had accidentally erased it.
if so, the best is just to re-install or repair your itunes in your computer.

You have to reset it, by holding down the menu button and the center button, then plug it in.
